In den Titelverbindungen liegen Derivate des Dopamins in schief ekliptischer Form sterisch fixiert vor. 5a, b und 6a, b erreichen nach intraperitonealer Applikation das Gehirn von Ratten und hemmen in Stammhirn- und Rindenarealen die Dopa- und 5-Hydroxytryptophansynthese. Gleichzeitig wird der Catecholamin- und Serotoninumsatz (5-HT) verlangsamt. 3-exo-(3,4-Dimethoxyphenyl)-N,N-dimethyl-bicyclo-[2.2.1]heptan-2-endo-amin (5a) hemmt in vivo die Wiederaufnahme von Serotonin in zentrale Neuronen in der Grosenordnung des Antidepressivums Imipramin. Neuropsychotropic Activity of Dopamine Analogous Bicyclo[2.2.1]heptanes The title compounds are derivatives of the neutrotransmitter dopamine, which show a sterically fixed ecliptic conformation. When applied intraperitoneally 5a, b und 6a, b reach the brain of rats and inhibit the synthesis of dopa and 5-hydroxytryptophan. The turnover of catecholamines and serotonin (5-HT) is decreased to a similar extent. The brain levels of the transmitters therefore remain fairly normal. The dopamine analogue 3-exo-(3,4-dimethoxyphenyl)-N,N-dimethylbicyclo[2.2.1]heptan-2-endo-amine (5a) inhibits the re-uptake of 5-HT into rat brain neurons to about the same degree as the antidepressant imipramine.
