these changes by day 7 of Vitamin D deficient diet. There was an increase of IL6, TNF, Elane and Nod2 but also there was a trend toward increased expression of IL17and Reg3g. Analysis of the microbiome showed a decrease in SFB and an increase in Clostrium Leptum and Strength 3 species. In invitro assays, Vitamin D alone intestinal epithelial cells pretreated with active Vitamin D alone had no effect on Lipocalin (Lcn2) expression. Interestingly as compare to IL17 treatment alone, Vitamin D in combination with IL17 led to increase the expression of Lcn2. There were not significant changes in the expression of Cxcl1, Csf3, Il6 and IL1. CONCLUSIONS: Our data suggest that mice under a Vitamin D deficient diet mice showed evidence of inflammation as measured by the expression of Il6, neutrophil elastase, and histopathology. From the in vitro assays, the data suggest that vitamin D can suppresses IL-17R mediated signaling as measured by IL-17 stimulated Lcn2 expression. Future work will identify mechanisms by which VDR effects IL-17R mediated gene expression.
