Serum levels of 26S proteasome are lover in patients with asthma

Introduction: Proteasomes - major nonlysosomal proteolytic complexes – participate in inflammation and in the pathogenesis of many diseases, including asthma. 26S proteasome is composed of three particles: 20S catalytic core and two 19S regulatory particles. It is shown that 20S proteasome concentration is elevated in blood serum in cancers and autoimmune diseases also in bronchoalveolar lavage from patients with acute respiratory distress syndrome (1). But there is no data about levels of 26S proteasome in association with diseases. Aims: The aim of this study was to evaluate serum concentration of 26S proteasome in Lithuanian patients with asthma and to compare it with healthy subjects. Methods: Patients with asthma (n=85, mean age 48.17±16.6yrs) and healthy subjects (n=74, mean age 39.54±16.19yrs) were involved in this study. Asthma was diagnosed according to GINA criteria. Peripheral blood serum was collected from all subjects and stored in -80°C. 26S proteasome concentration was measured by enzyme-linked immunoabsorbent assay method with Human 26-PSM ELISA Kit (Elabscience, China) according to manufacturer9s protocol. Results: We found higher 26S proteasome serum levels in healthy controls when compared with asthma group (3.61±3.8 ng/ml vs 2.49 ±4.2 ng/ml, p Conclusions: This study showed lower serum levels of 26S proteasome in patients with asthma.
