Amniotic membrane loaded with bone marrow mesenchymal stem cells facilitates the healing of deep burn wound

The amniotic membranes have unique properties including transparency, stimulatory effect on cell proliferation and lack of immunogenicity, which may have clinical utility for cell delivery. In this chapter, we investigate the possibility of amniotic membranes loaded with bone marrow mesenchymal stem cells for healing of deep burn wound. The mesenchymal stem cells derived from human bone marrow were harvested, purified and cultured by combining density gradient centrifugation with adhering method. And then these cultured cells were seeded and bred on the stroma surface of the amniotic membrane to form the engineered skin. The biological attachment and growth of cultured mesenchymal stem cells were observed. Ten cases of deep burn wounds were applied with the engineered skin. The results showed that the mesenchymal stem cells adhered to the surface of amniotic membrane quickly after being seeded. The wounds treated with engineered skin healed rapidly with good clinical “take”. The average time of wound closure in engineered skin group was shortened than that of control group. There were no obvious evidence of immunological rejection and inflammatory reaction during the observation. It suggested that the amniotic membrane loaded with bone marrow mesenchymal stem cells may be an excellent tissue engineered skin for healing of deep burn wound.
