Growth Conditions and Genetic Characterization of a New Manganese Resistant Strain

Phytostabilization, the use of plants for insitu stabilization of tailings and metal contaminants, is a feasible alternative to costly remediation practices. Ecological adaptation of metal contamination is significantly enhanced through plants inoculated with plant growth-promoting bacterial. Rhizosphere soils of Chenopodiumalbum Linn were collected by means of field surveys of tolerating plants which grew in Xiangtan manganese tailings.1 light-gray manganese-resistant strain numbered XY was isolated from the rhizosphere of fresh plant through concentration gradient screening technique. The cellular form, physiological and biochemical tests were conducted, as the growth curve and effects of temperature, pH, NaCl concentration to growth of the strain tested were investigated. The results of the tests showed that 1) the strain reached logarithmic phase in 12-24h; 2) the optimum temperature range of the strain was 35隆æ-40隆æ 3) the optimum pH range of the strain was 6.8-7.3, 4) the optimum NaCl concentration range of the strain was 5g/L. The status of the strain in the taxonomy and phylogeny has been conformed according to 16S rDNA sequences, the strain XY belongs to Arthrobacter genus.
