Resveratrol production from Polygonum cuspidatum by one-pot green extraction and bioprocessing of polydatin

Polygonum cuspidatum is extensively utilized globally due to its abundant content of stilbene compounds in plant cells. In this study, we developed a one -pot green extraction procedure in situ that combined enzymatic hydrolysis of polydatin with extraction of resveratrol using a Deep Eutectic Solvent (DES) from Polygonum cuspidatum. Fifteen DESs were synthesized, consisting of choline-alcohols (n = 3), choline-organic acids (n = 5), choline-amines (n = 2), and choline-sugars (n = 5), and their productivity in combination with three types of enzymes were evaluated. Based on the total yield of resveratrol and polydatin and conversion rate of polydatin, beta-glucosidase assisted choline chloride -glycol DES was selected. To maximize the yield of resveratrol, singlefactor tests combined with response surface methodology were selected to optimize the extraction process. Under optimal extraction conditions, the extraction efficiency and yield of resveratrol were 99.74 % and 85.56 +/- 0.96 mu mol/g (19.53 +/- 0.22 mg/g, n = 3), respectively. Additionally, beta-glucosidase can still keep excellent enzyme activity in choline chloride -glycol DES at 64 degrees C with the 100 % of conversion rate of polydatin. This study offers a new, cost-effective, efficient, and environmentally friendly method for enhancing resveratrol production. Additionally, it showcases the applicability of enzyme -catalyzed extraction utilizing DES as a green and sustainable technique for available resveratrol from Polygonum cuspidatum.
