Multifunctional group mediated double cross-linked polysaccharide complex hydrogel for microenvironmental regulation and repair of endometrial injury

Endometrial injury can cause inflammation, leading to endometrial cell growth restriction, accompanied by fibrous tissue hyperplasia and adhesion in severe cases. Traditional treatments have problems such as high adherence rate, immunogenicity and intimal fibrosis. To solve the above problems, a multifunctional group mediated double cross-linked polysaccharide complex hydrogel was designed. Multiple groups were introduced into the hydrogel system by self-polymerization and amidation, and multi-network SA/PDA/CMCS-Arg injectable hydrogel was formed by non-toxic ion triggering system. The microenvironment can be regulated through the triple regulation of fibrinogen capture, active oxygen scavenging and bacteriostasis mediated by multifunctional groups in the inflammatory environment, so as to reduce the degree of tissue fibrosis and inflammatory reaction, and realize the repair of endometrial tissue. The effect of hydrogel on repairing endometrium was verified through Mouse endometrial injury model in vivo. The experimental results showed the composite hydrogel has a more suitable structure and performance for endometrial repair. After 7 days of hydrogel treatment, the matrix, epithelium, and glands of the damaged endometrium recovered well, and the degree of fibrosis was much lower than that of the control group. These results indicate that SA/PDA/CMCS-Arg injectable hydrogel has the potential to be an alternative material for endometrial repair.
