•Dehydroevodiamine, a quinazoline alkaloid, exhibits potent antitumor efficacy and low toxicity in pancreatic cancer.•Dehydroevodiamine suppresses tumor cell stemness through DDIT3/TRIB3/AKT/mTOR pathway.•Dehydroevodiamine is a novel DDIT3 activator, and activation of DDIT3 could be a promising therapeutic strategy for treating pancreatic cancer.
