•The study shows a molecular basis of interaction between Spike and TLR4 of wild type and significant SARS-CoV-2 variants.•It shows Omicron Spike strongly interact with the TLR4 compared to wild-type, Alpha, and Delta variants.•H-bond formation during Spike and TLR4 interaction in wild type, Alpha, Delta, and Omicron.
