•Physical exercise may enhance GLP-1 secretion by interleukin-6 action.•No mathematical model described IL-6-induced GLP-1 secretion due to physical exercise.•We propose a mathematical model of IL-6/GLP-1 interaction following moderate exercise.•We defined the sensitivity of GLP-1 secretion to exercise-mediated IL-6 action.•Model parameters are computable in the single subject, thus have clinical potential.
