•Fe3O4-CaO2-PDA self-supplies ·OH and depletes GSH while producing oxygen.•Fe3O4-CaO2-PDA has dual regulatory functions.•PTT can accelerate the cascade reaction of Fe3O4-CaO2-PDA hybrid nanozyme.•PTT-enhanced Fe3O4-CaO2-PDA hybrid nanozyme has excellent biofilm removal ability.•Fe3O4-CaO2-PDA hybrid nanozyme has a role in modulating inflammation in mice.
