•Mangiferin attenuates osteoporosis in ovariectomy or iron dextran-induced mice.•Mangiferin prevents ferroptosis in ovariectomy or iron dextran-induced mice.•Mangiferin inhibits ferroptosis in iron-dextran or erastin-induced osteoblasts.•Mangiferin activates SLC7A11/GPX4 pathway by directly bonding to Keap1.•Knockdown of Nrf2, SLC7A11 or GPX4 abolishes the protective effects of mangiferin.
