Distinction between multiple primary lung cancer (MPLC) and intrapulmonary metastasis (IPM) holds clinical significance in staging, therapeutic intervention and prognosis assessment. However, it poses significant challenges, particularly for common histologic subtypes. The partnering of molecular profiling and clinicopathologic data might increase the reliability of lineage definition. Here, we proposed a multi-omics algorithm that delineates spatiotemporal heterogeneity through genomic, transcriptomic and cellular characterization, which might assist in lineage calling in multifocal lung cancer, surpassing the limitations of clinicopathologic classification.
