HPCE monitoring of phosphorylated histone H1 variants

This chapter summarizes the effects of buffer concentration and buffer composition on analysis of phosphorylated histone H1 variants by High performance capillary electrophoresis (HPCE) using the dynamic coating agent HPMC. HPCE continues to become more widely used as a technique for the separation of peptides and proteins. The objectives of the work described in this paper were to investigate the effects of different buffer concentrations and compositions, respectively, on the elution order and separation of H1 histone subtypes and their phosphorylated modifications isolated from two different mammalian species. HPCE, combining the advantages of conventional gel electrophoresis and high performance liquid chromatography, may offer substantial improvements over other analytical techniques. The aim of the first studies has been to determine whether HPCE can be used for the simultaneous separation of both rat testis histone H1 subtypes and their different phosphorylated forms.
