Hemoglobin Assisted Synthesis of Au NCs@MBA-TPP Nanoadapters for Mitochondria Targeted Multimodal Combined Therapy

The complexities of light penetration, short O-1(2) life, severe tumor hypoxia, overexpressed reductiveglutathione(GSH), and acidic conditions in the tumor microenvironment (TME) limitthe success of photobased multimodal combined therapy (MCT) in anticancer.Herein, the mitochondria targeting nanoadapters with multistimulusresponsibility are fabricated for the photothermal imaging guidedMCT. The obtained Au NCs@MBA-TPP NAPs can be triggered simultaneouslyby overexpressed H2O2 and GSH in TME, as wellas near-infrared (NIR) laser, achieving chemodynamic therapy (CDT),NIR motivated photothermal therapy (PTT) and photodynamic therapy(PDT). Furthermore, the hematosin in Au NCs@MBA-TPP NAPs causes O-2-generation and GSH depletion for the enhancement of PDT andCDT with the mitochondria targeting enhanced MCT effectively by thephotothermal imaging guide. Hence, our design provides a new strategyfor the integration of diagnosis and treatment with TME modulationto potentially improve the MCT in tumor treatment potentially.
