093 Atopic dermatitis (AD) and psoriasis overlap: A new disease subset of AD defined by spatial molecular analysis of AD and psoriasis

Atopic dermatitis (AD) is a chronic inflammatory skin disease with heterogenous endotypes. Among T cell-mediated inflammatory skin diseases, psoriasis has been thought to be a prototypic Th17 cell-dependent disease, and AD is considered a representative Th2 immune response. According to the research, Asian AD patients had a combination of AD and psoriasis features. Studies showed that Korean AD patients exhibit Th17 markers as well as Th2 features when compared to European-American patients. In order to contrast these psoriasis-like AD skin lesions with typical AD and psoriasis, we profiled whole gene expression levels in each lesion thanks to spatial transcriptomic technology. Using the GeoMx®, a commercially accessible spatial transcriptomic platform, four individuals with prototypic AD, six with overlapped clinical and histological characteristics, and four with typical psoriasis were investigated. We confirmed the molecular spectrum of diseases from AD to psoriasis with the genes such as CCL24, CCL26, IL13, and IL36G. Thereafter, our results may contribute to the strategies for improving the diagnosis and treatment of inflammatory skin diseases.
