•The Equine β-defensin-1 (eBD-1) promotes cytokines mRNA expression in S. aureus-infected macrophages regulated by PI-3K-NF-κB pathway.•The eBD-1 increases macrophages phagocytose S. aureus associated with FAK-paxillin signalling pathway.•The eBD-1 has a potential application ability in alternatives to antibiotics.
