•A peroxidase-dead PRDX1 variant mouse strain was generated by mutating its peroxidatic Cys at 52 (Cys52) to Ser (PRDX1Cys52Ser).•PRDX1Cys52Ser mice were resistant to WD-induced NASH and liver fibrosis.•PRDX1Cys52Ser mice showed significantly reduced NF-kB and STAT1 signaling activities and their relevant pathways in the liver.•PRDX1 Cys52Ser variant also conferred resistance to MCD-induced NASH by suppressing NF-kB and STAT1 signaling activities.
