•The main polyphenols of pomegranate peels were identified by LC-MS/MS.•Punicalagin from pomegranate peels was extracted under optimal UAE.•Punicalagin was purified by D101 macroporous resin and prep-HPLC.•α-Glucosidase inhibitory mechanism of punicalagin was studied.•Molecular docking indicated the interactions between punicalagin and α-glucosidase.
