•Low BCF values indicate that HFPO-DA does not readily bioaccumulate in flies.•Lifespan was significantly reduced starting at 102 mg/kg/day in both sexes.•Exposure-induced locomotion score decline was more evident in females than males.•Brain gene expression patterns are specific to sex and exposure duration.
