•Germination and enzymatic hydrolysis increased the antioxidant potential of mustard grains.•14 peptides and 17 phenolic compounds were detected by UPLC-HRMS analysis.•Dipeptides and phenolics release after bioprocesses may have contributed to the antioxidant potential improvement.•Germination and enzymatic hydrolysis increased Caco-2 cytotoxicity indicating a potential antitumor activity.
