Discovery of pyrido[4,3-d]pyrimidinone derivatives as novel Wee1 inhibitors

•Novel Pyrido[2,3-d]pyrimidin-4(1H)-one derivatives were discovered as potential Wee1 inhibitors.•The optimal compound effectively inhibited Wee1 and demonstrated robust antiproliferative suppression in MV-4-11 cells.•The optimal compound exhibited favorable stability in mouse liver microsomes in vitro.
