•Defective copper-based metal organic frameworks (Cu-MOFs) were synthesized.•d-SPE technique was employed for enrichment of organophosphorus pesticide (OP).•Defective Cu-MOFs showed excellent adsorption kinetics and capacities for OPs.•A sensitive determination method for OPs based on defective Cu-MOF was developed.•The OPs adsorption mechanism on Cu-MOFs was validated via the DFT calculation.
