Duplex real-time qPCR for Feline Herpesvirus-Ⅰ and Feline Parvovirus

Abstract To establish a rapid detection method for feline herpesvirus-Ⅰ (FHV-Ⅰ) and feline parvovirus (FPV), the specific primers and probes were designed according to the gB gene and VP1 gene of the highly conserved regions in FHV-Ⅰ and FPV strain. We optimized the reaction system and conditions of primers and probes, and a dual real-time fluorescent qPCR method that can simultaneously detect the above two viruses was successfully established. The standard curve of the method has an excellent linear relationship, and the correlation coefficient (R2) is above 0.99; the amplification efficiency is between 90% and 110%; the minimum detection limit is about 20 copies/μL; the coefficient of variation within and between groups is less than 2% And only positive for FHV-Ⅰ and FPV gene amplification, but negative for FCV, SVV, VSV, and other infectious disease-related pathogen amplification.
