Inflammasome implications in metabolic disorders

Growing evidence highlights the relevance of inflammasomes, including nucleotide-binding oligomerization domain leucine-rich repeat and pyrin domain-containing protein 3 (NLRP3), absent in melanoma 2 (AIM2), NLRP6 and NLRP1 as pivotal players in the pathophysiology of metabolic disorders, including obesity and type 2 diabetes mellitus (T2DM) and related comorbidities as well as atherosclerosis and nonalcoholic fatty liver disease (NAFLD). In this regard, intensive research efforts both in humans and in animal models were focused on the characterization of the pathophysiological role of inflammasome signaling in metabolic diseases, providing novel insights about the development of novel therapeutic strategies aimed at undermining inflammasome-induced immune-metabolic alterations in such disorders.
