•Inherited protein S (PS) deficiency is an established risk factor for venous thromboembolism.•The impact of missense mutations and their location on the risk of thrombosis is not clear.•Mutations located in a specific region of PS may have a higher impact on thrombotic risk.•Thus, genetic analysis could contribute to further risk prediction in PS-deficient patients.
