PDF file - 59K, In this model, nitric Oxide regulates mTOCR1 activation by acting on the TSC1/2 complex through reversible nitrosylation of TSC-2, which blocks TSC1/2 dimerization. In the absence of NO, the TSC1/2 complex suppresses activity of the Rheb GTP-ase, which activates mTORC1 in GTP-bound form. In the presence of NO, the TSC1/2 complex does not form, and Rheb remains active and capable of activating mTORC1 and the downstream mTOR pathway.
