•Cooperatively TGF-β blockade to reverse immunosuppression post PDT.•PDT-induced CD44 expression was used for incremental tumor targeting efficiency.•PDT-generated ROS was exploited for controlled drug release.•HA-aldehyde-monosubstituted β-CD to minimize the impact on its binding with CD44.•BzPGA with strong π-π stacking force improved the encapsulation efficiency of LY.
