•A split-type-based cathodic amplification strategy was designed in bipolar ECL.•The anodic ECL responses were regulated by the conductivity of the cathode.•The work effectively avoids the reduced light transmittance or protein activity.•The bipolar ECL sensor exhibits high sensitivity and good selectivity.
