•UV–visible study confirmed that APBBO derivatives interact with BSA.•Fluorescence study confirms static quenching and binding.•Docking studies along and thermodynamics confirmed H-bonding and hydrophobic interactions are the main driving forces.•CD spectra showed post binding secondary structural changes in BSA.•APBBO derivatives altered the esterase-like activity of BSA.
