Effect of Cistanche deserticola Polysaccharide on Learning and Memory Ability and Synaptic Protein Psd-95 Expression in Alzheimer's Disease Rats

The aim was to study the effect of Cystanche deserticola polysaccharide on learning memory and synapse-related protein PSD-95 expression in the Alzheimer's disease (AD) rat model. The AD model rats were randomly divided into control group and models group and large, medium and small dose of C. deserti-cola polysaccharide groups (50, 25, and 12.5 g/kg for day), 15 patients in each group were continuously infused for 4 months. Fifteen SD rats were injected with artificial cerebrospinal fluid (CSF) in the bilateral lateral ventricles as control groups. After the last gavment, platform test and MORRIS water maze the changes of PSD-95 protein were determined by immunofluorescence, and PSD-95mRNA expression level by RT-PCR. The number of platform crossing was significantly reduced in the model group (p < 0.01), and in the large, medium and small dose groups compared to the model group. Immunofluorescence and RT-PCR showed that the polysaccharide can upregulate the expression of PSD-95 protein and mRNA in rat hippocampus (p < 0.05). C. deserticola polysaccharide can improve the learning and memory ability of AD model, including increas e PSD-95 protein expression to promote the synaptic transmission function.
