Invariant natural killer T (iNKT) cells are a subset of innate-like T cells with great potential for developing cancer immunotherapies, including approaches based on chimeric antigen receptors (CAR). In this issue, Ngai and colleagues report that the transcription factor lymphoid enhancer-binding factor 1 (LEF1) optimizes functional properties of iNKT cells that promote antitumor immunity, raising enthusiasm for the development of robust cancer immunotherapies based on CAR-modified iNKT cells.
