Surface Plasmon Resonance Sensor Based on Kretschmann Configuration for Dengue Virus Detection

Surface plasmon resonance (SPR) has been extensively used as a diagnostic tool for virus and bacteria detection (biosensors). This research detected dengue virus (DENV) using a non-structural-1 monoclonal antibody (NS-1 mAb) on the gold/EDC-NHS/IgG component of SPR. The experiment was conducted in real-time by monitoring the SPR angle and response unit (RU) change. As a result, it was observed that the presence of the functional molecule on the SPR chip caused the shift of the SPR angle to the higher incident angle, indicating a refractive index change proportional to the attached molecule layer. The increase of RU value due to the injection of the biomolecule into the SPR chip represents the SPR angle shift. The result revealed the potential of the SPR as a promising tool for DENV detection.
