•We report the reduction of Cu(II) to Cu(I) of CuAl-LDH nanosheets by glutathione.•The generated Cu(I) can serve as a catalyst to catalyze the bioorthogonal reaction.•The generated Cu(I) can catalyze the Fenton-like reaction for chemodynamic therapy.•The CuAl-LDHs can achieve precise and efficient elimination of tumors in vivo.
