•Manuscript clarifies chelating ability of carnosine towards Cu2+, Mn2+ and Zn2+, combining potentiometry, NMR and MS.•Potentiometry allowed to define speciation models and formation constants at different temperature and ionic strength.•1H NMR spectroscopy further confirmed the speciation model for Zn2+ -carnosine system.•High Resolution (HR) Matrix Assisted Laser Desorption Ionization (MALDI) Mass Spectrometry (MS) clarified chelating mode.•Sequestering ability of carnosine towards Cu2+, Mn2+ and Zn2+ was quantified in physiological conditions.
