Simultaneous extraction and quantification of circulating mitochondrial and nuclear DNA using a single plasma sample to predict specific molecular diagnostic implications

•Our study reports a reliable, cost-effective, and highly sensitive approach for the simultaneous extraction of both cir-mtDNA and cir-ncDNA using a single aliquot of 300 µL plasma sample.•This approach may help to avoid sample volume issues and the use of costly commercial kits, and kits and decreases the time and efforts in downstream diagnostic processes.•The cir-DNAs extracted using described approach are suitable for downstream analysis and provide a more precise and valuable tool to discriminate the significance of cir-mtDNA from cir-ncDNA in disease diagnosis.
