•TGF-β1 and MCAO/R injury upregulated BRD4 expression in vitro and in vivo.•Inhibition of BRD4 decreased TGF-β1-induced meningeal fibroblast activation.•BRD4 knockdown inhibited fibrous scarring and improved neurological function.•Inhibition of BRD4 decreased the phosphorylation of Smad2/3 in vitro and in vivo.
