•When PLA/PEG/Chitosan/FA/DNA nanoparticles interact with MCF-7 cells, their loading percentage and biocompatibility improve.•Physicochemical properties of nanoparticles for properly targeted transfer to MCF-7 cells.•An increase in Chitosan-FA percentage on gene transfer efficiency was seen positively by PLA/PEG/Chitosan/FA/DNA nanoparticles.
