•Two novel cocrystals of acetazolamide formed with two bipyridine compounds.•Single-crystal X-ray diffraction reveals layered and cavity structures.•Simultaneously improve solubility and permeability of acetazolamide.•Cavity structure in crystal lattice causes better water solubility and permeability.•New insight for the application of N-containing compounds in cocrystallization.
