•Smyd2 is the most prominently expressed Smyd mRNA in human bone tissue and mouse osteoblasts.•Smyd2 is a lysine methyltransferase that modifies both histone and non-histone proteins.•Smyd2 depletion stimulates osteoblast proliferation in MC3T3 cells.•Smyd2 loss increases Ezh2 protein expression.•Loss of both Smyd2 and Ezh2 promotes osteoblast mineralization in MC3T3 cells.
