•Uniform agarose microspheres were prepared using membrane emulsification method.•The average pore size decreased using dextran with different molecular weights.•The average pore size varied according to different crosslinking procedures.•The separation range was regulated by both dextran-grafting and crosslinking.•The pressure-resistance was improved by both dextran-grafting and crosslinking.
