Mechanistic efficacy assessment of selected novel methanimine derivatives against vincristine induced Neuropathy: In-vivo, Ex-vivo and In-silico correlates

•Novel methanimine derivatives (KB9 and KB10) were tested for in-vivo, ex-vivo and in-silico analysis.•During in-vivo studies, both compounds enhanced PWL, PWD, TWL and paw licking and flinching in their respective tests.•In ex-vivo tests, KB9 and KB10 blocked oxidative stress markers, both decreased the TNF-α and IL-1β during ELISA. NF-κB and COX-2 levels were reduced in immunohistochemistry and during western Blot analysis they effectively reduced NF-κB, TNF-α, IL-1β, and COX-2 expression levels.•During in-silico analysis, both test compounds showed preferred affinity for NF-κB and COX-2 target sites.•KB9 and KB10 may be therapeutically effective in management of neuroinflammation, inflammatory and neuropathic pain.
