•Phosphatase adsorption process onto nanoclay was spontaneous and enthalpy driven.•Immobilized acid phosphatase had significantly lower Vmax than free enzymes.•Alkaline phosphatase when immobilized had significantly higher Vmax than free enzyme.•Poorly ordered aluminosilicates in nanoclay altered both enzymes' substrate affinity.
