Quantification of prefusion conformation for HIV vaccine using size-exclusion chromatography

•A novel, cost-effective SEC method is developed to quantitate HIV prefusion conformation during production of vaccines.•Conformation specific calibrator for % prefusion quantitation is not required for analysis.•Conformation specific antibodies can be screened using this method for further HIV clinical candidate development.•Molecule and reagent quality can be monitored in one assay.•This method can be applied to other molecules that have prefusion conformations, through use of appropriate binding partners.
