The available zebrafish genotyping protocols do not provide the most accurate results, mainly due to the use of electrophoresis gels. Single nucleotide polymorphisms, insertions, deletions and point mutations are most accurately detected by sequencing. Here, we describe a high accuracy and scalable single-day protocol to detect zebrafish mutants by Sanger sequencing. This protocol can be used to successfully genotype zebrafish adults and larvae.
