•The damage diphenoxylate-caused was more serious than loperamide did in organs of mice.•Diphenoxylate-induced constipation model shown the more adverse environment of intestinal microflora compared with loperamide did.•Mulberry ferment exhibited a strong laxative effect under two types of models.
