•The randomized, crossover trial design has advantage of strong causal inference.•First study evaluated genome-wide mRNA following ozone exposure using RNA-seq.•Ozone exposure led to significant dysregulation of circulating transcriptome.•Affected mRNA plays important roles in various cardiopulmonary diseases.
