•Mannose binding NTL-125 lectin inhibits the virus replication in Vero cell line•Native and mutant spike proteins show higher binding affinity to NTL-125 than the corresponding Spike-hACE2•Higher affinity of NTL-125 to RBD competitively inhibits Spike-hACE2 interaction.•NTL-125 disrupts protective viral shield by interacting with glycan moieties of spike•NTL-125 could be promising therapeutic molecule and or biosensor for COVID-19 detection.
