•Spinal projections to the lateral parabrachial nucleus (LPBN) are glutamatergic.•Periaqueductal grey matter (PAG)-LPBN projections are glutamatergic and GABAergic.•Opioids reduce synaptic transmission at spinal and both PAG-LPBN synapses.•Cannabinoids reduce excitatory and inhibitory PAG-LPBN synaptic transmission.•Benzodiazepines enhance inhibitory transmission at PAG-LPBN synapses.
