•The binding stereospecificity of stipuol enantiomers with human serum albumin (HSA) were compared.•(R)-Stipuol binds HSA stronger than (S)-stipuol.•(R)- and (S)-stipuol bind site Ⅱ of HSA.•Binding of stipuol enantiomers induces HSA conformational changes.
